I won it from going up and down the east coast. The pool respawn times are really quick so don't be afraid to turn around and go back in the direction that you just came from.
Also, I'm sure you know this but obviously set your hearth to BB, get walkerwalking elixirs or a dk in your group if you aren't a dk, priest or shaman. Swiftness potions REALLY helped me.

Be prepared is all I can say.
The advice I got from this site was invaluable in helping me win the first time I entered.
1) set hearth to BB
2) Get into position early (20 minutes at least)
3) one minute before the event starts put on your sharpened hooks, aquadynamic fish attractors...whatever. Also have your water walking pots in effect.
4) Have fish finding onI tried to leave one tastyfish in each pool before I moved on to the next spawn point. My rationale was that others would see the pool, stop to fish, and only get one fish from the pool, making it less time-effective for others, while I was moving along down the coast.

I won the Tournament today, after months of trying, and I can only add one thing to what Gimeg said. Never give up!
For the past month, I've had a horrid streak of luck, often landing only 32-34 Tastyfish by the time the winner was announced. I knew I was doing everything right, and that it was just the RNG hating on me, though. :)
Today, fortune was definitely in my corner. I fish on Yojamba Isle, off the very Northwest tip of STV, and there was only one other angler there, today. Pools kept appearing practically at my feet, and I caught very little trash. I was practically hyperventilating as I hit the hearthstone, and couldn't believe my eyes when I saw nobody between me and Riggle Bassbait!
Just make sure you read everything on this site, and then keep at it! You'll eventually win the thing! Thank you El, and everyone who posts here! You all do a great service to everyone who fishes in WoW.

This past Sun. (the 28th) I came closer than ever to winning. I was beat literally by a few steps. It was a good day to win. There didn't seem to be as many as usual competing, and I didn't get ganked or even attacked at all.
Thinking back at what I did wrong;
#1. I forgot to set my hearth. Had to ride back.
#2. I got spooked by a splash and clicked my bobber to soon. (one time)
#3. I, several times clicked to fast trying to get my bobber in the pool, and messed up a good cast.
#4. A couple of time's I took a chance on a bobber very close to the edge of the pool, caught junk.Lesson learned, don't get in hurry when casting, don't forget to set the hearth, and get it in the pool. Any of the things above not done, and I would have won.

I fished on the "Twink Island" aka the troll one in northwestern stranglethorn, it worked fine for me and another fisher who suggested it to me, Ive also gotton 38/39 on that island when the winner was announced so its lucky (at least for me) just remember to set your hearth at bb.

If you have gotten to 10 or less fish needed and no winner has been called, consider using swim speed or swiftness potion to get to the last few pools as it's faster than mounting and riding.
Also, have your prize picked out (pole or hook) before you hand in the fish and click it immediately as others may be arriving just behind you.

These might be listed in the "well, duuuh" section of advice, but... :)
1 - Get your fishing level as high as possible to avoid grays. Lures, fish hooks, hats, enchants etc.
2 - Set your hearthstone to Booty Bay so you don't have to ride back when you have all your fishies
3 - Find a area where there are few other players. I like the eastern coast.
4 - If possible, bring a friend with you that is not participating in the contest. A bodyguard! :) This will make it less likely that enemy players gank you, and will also prevent others from fishing in the same pool as you. "Ohh, there are already two players by that one, I better find another pool".
5 - Have fish finding on. :P

I just won the contest in 13 minutes today, and I think I have a couple of tips that helped me. I may be way off, but:
Aside from the reiterated hearth setting, and water walking, i made sure to
-eat some food with a haste rating buff (dunno if that helped),
-set up as much as i possibly can ahead of time with the fish tracker, and attaching the lure,
-reprogram my hotkeys
-1 fish
-2 aspect of the cheetah (i'm a hunter and that helps my unmounted movement speed)
-3 mount
-4 elixir of water walking
-5 hearthstone (set to booty bay).
-I also used a Glow Worm lure because it lasts an hour and I can attach it early in preparation, and
-Another thing that helped me cast faster was using a pole with a visible string. I had been using my Bone Fishing Pole that I got from Dalaran dailies, but there is no visible line so I didn't know if I had a bad cast until the bobber started to appear. Switching back to my Nat Pagle's Lucky Fishing Pole helped immensely!!!As for winning in 13 minutes, that was only because it was a holiday weekend (fewer contestants) and not a single person on the east coast, but me. XD

I've won two of the past four contests on my server, using the same tactic each time. My thanks go out to the helpful folks at El's Anglin--lots of good info on this site!
I'll reiterate what other folks have said:
1. Set hearth stone to Booty Bay
2. Use elixir of water walking if you don't have a way to cast it on yourself; even water form or levitate is slow compared to riding along the water on your ...
3. Epic mount.
4. Scope out where pools spawn, and what other players do, and do whatever lets you fish unimpeded for the longest amount of time. On my server, people seem start either at the bottom or the top of the western coast and fish their way down or up. I start in the middle of the western coast. There's a spot between Grom'gol and Gurubashi Arena with some little islets jutting off of the coast. If you start on one of the islets, when the contest starts, you can see as many as four or five pools at once on your ...
5. Track Fish.
6. Don't fight with others for fish. My spot on my server lets me fish without seeing other would-be master anglers for around six or seven minutes--plenty of time to catch my first twenty tastyfish or so. The high density of pools in the spot I start in means I can spent a lot of time fishing and little time running from pool to pool. However, when other people come by and start fishing in a pool I'm already fishing, I hightail it to another pool. I'm better off to fish unimpeded in my own pool than to fight with others for one or two tastyfish, and then run to the next pool with them to keep fighting. Both times I won I even abandoned a pool when I was at 39 tastyfish to run to an unoccupied pool--but that may not have been the most ideal move.
7. Don't hope your bobber is in the pool. Recast if you're not sure if your bobber is in the pool. I got burned on that a couple of times in my last contest--I believe Tastyfish pools are less forgiving than the Northrend pools I had been fishing!
8. Don't cast too fast after clicking the bobber to loot--erring on the side of cautious when your nerves are jittery with the contest anyway is better than losing a tastyfish, fifteen seconds of casting, and letting the competition get ever closer.
9. Practice your fishing. This probably goes without saying, but if it takes you two or three casts on average to get your bobber in a pool, you'll save tons of time compared to somebody who takes five or more. Also, it helps to have a base fishing skill high enough that you don't have to worry about lures.
10. Don't give up if you have bad luck! Both times I went on to win, I made some of the mistakes I mentioned above (multiple times!), and I was sure would cost me the contest. Everyone else might be having bad luck too.I also made good use of macros--I have one that casts multiple times with one button press, which means I can fish closer to the coastline if I'm standing on the water fishing toward the sand, which is how I prefer it. I'm more used to fishing that way with levitate, as I'm a priest, so if you're more comfortable fishing from land, you might want to stick with your usual style. The advantage of the multicast macro is it will recast if I get a "water too shallow" or similar error message, but it won't move the bobber if I get a message like that after I've already cast.
Another of my macros is my hearthstone, but I've set it up to show how many tastyfish I have in my bags rather than just my hearthstone. It just saves some space on the bar, but is probably not necessary. I also put my mount and my elixir of water walking on the same bar as my fishing skills, just so I have ready access to them while I'm fishing.
Skill plays a huge role in this contest, but luck helps a lot too. It's really hard to win when you catch several firefin snapper in a row. If you do everything right, you can still lose if you're up against somebody who doesn't quite fish as well but simply has great luck. Just keep trying, and eventually your skill and your luck will align, and you'll be holding either an amazing fishing pole or a fun fish-making trinket. Or both!

It's a combination of luck and practice. I know no one wants to hear that but it's true. Having one is no good at all without the other. So yes, be sure to use all the tips you read on these forums, I consider them to be sorta assumed requirements. Even once you've read all the tips and you're properly prepared, it still takes persistence--lots (lots) of losing tries before you have enough practice to make good use of the luck when it finally comes your way. This is a contest where even the tiniest finesses and sharp instincts can make a difference, and lots of practice is the only way to get 'em. Nothing you can read on a forum will make up for that.
Edit For Update: I participated in the contest again on a different character this past weekend. I was fresh off my 4th win in the previous week and went in with every bit of preparation, skill, equipment, adrenaline, and confidence. And I lost in the most spectacular fashion--only 22 tastyfish when the winner was called, and a whopping 15 trash items (oily blackmouth, firefin snapper, and stranglekelp). I fished, or at least attempted to fish, in the same area where I usually do, but it just plain didn't work out for me this week. Even though I had disappointing luck right out of the gate, I still gave it every bit of my all until the moment the winner was called. Further evidence to show that there is no "trick" or "advice" that will help. It doesn't mean you're doing anything wrong, or that there's anything you can do better, just gotta keep trying the best you can.
